have you tried sudo from the commandline?

or even open up a xterm and do a gksudo synaptic, that way you will
see the errors, (hopefully).

if that doesn't help, I would try creating a new user... with the gui
tool, it will allow you to make the user part of the appropriate
groups so that it can sudo, then switch to that user and try the same
thing.
