I don't know, but I suspect it would ... if the HD copy is basically just a
dd of the cd (i.e. copied the /etc/shadow with the scrambled password), then
it should function the same. 

When running from the cd/dvd, the modified (modifiable) files are stored in
ramdisk.

Does this apply to Knoppix that has been loaded to the HD, versus running
from a CD or a DVD?
 
  The reason I ask is that I'm told that Knoppix does not use the HD, while
running from CD or DVD, unless a swap drive is already defined on the HD.
